On July 20, 1944, Pierce was born in Wellington, Texas. She grew up in Lubbock and attended high school there with Butch Hancock and Joe Ely. In the 1980s, she started writing songs after receiving encouragement from Ely and David Halley.

The compilation album Across the Great Divide: Songs of Jo Carol Pierce, with 19 renditions of her songs, was created in 1993 by her fellow Austinites in the music industry. She has made other cabaret pieces besides her CDs, including Bad Girls Upset by the Truth, a semi-autobiographical musical comedy, and In the West. It received its world premiere at SXSW in March 1993 and was eventually transformed into her 1996 debut solo album of the same name. She self-released Dog of Love, her second solo album, in 2008.

Her high school lover, Jimmie Dale Gilmore and Pierce were married in 1963; they had a child together before divorcing in 1967.
